Description Vector: Lambda Zap; Medicago truncatula plants were infested with 300 spotted-aphids each. Tissue samples were collected by harvesting the entire shoots from 13 plants after 12, 24, 48, 72 and 96 hours of infestation. Total RNA was extracted from each sampling time and equal amounts of total RNA were pooled together for mRNA purification for cDNA library construction. The cDNA was directionally ligated into the Uni-Zap XR vector (Stratagene) and packaged using the Gigapack III Gold packaging extracts. Phagemids containing cDNA inserts were in vivo excised from the recombinant Uni-ZAP XR vector using ExAssist helper phage and the E. coli strain XL1-Blue MRF' (Stratagene). Excised plasmids were plated using SOLR cells.
