Vector: pBluescript SK +/-; Site_1: EcoRI; Site_2: XhoI; cDNA was prepared from polyA+ enriched RNA from the mixture of cotyledons of five days old plants and leaves of two weeks old plants. The cDNA was directionally ligated into the Uni-ZAP XR vector from Stratagene and packaged using Gigapack III Gold packaging extracts. Plasmids containing cDNA inserts were excised from the recombinant lambda-ZAP phage using Ex-Assist helper phage and propagated in SOLR cells. |
