Description Vector: pBluescript SK-; Site_1: EcoRI; Site_2: XhoI; At the trifoliate stage, M. truncatula plants were transplanted to phosphate-free sand and grown for a further 30 days. During this period, they were fertilized twice weekly with 1/2 Hoaglands solutions containing 20uM potassium phosphate. cDNA was prepared from polyA+ enriched RNA. The cDNA was directionally ligated into the Unizap XR vector from Stratagene and packaged using Gigapack III Gold packaging extracts. Plasmids containing cDNA inserts were excised from the recombinant lambda-Zap phage using Ex-assist helper phage and propagated in XLOLR cells.
