Description Vector: pBluescript SK-; Site_1: EcoRI; Site_2: XhoI; Immature pods, ranging in age from 15 to 30 days after pollination, were collected from greenhouse-grown plants. At harvest, seeds were removed from pods and isolated pod walls were collected and immediately frozen in liquid nitrogen. Pod walls were pooled for mRNA extraction. cDNA was prepared from polyA+ enriched RNA. The cDNA was directionally ligated into the Unizap XR vector from Stratagene and packaged using Gigapack III Gold packaging extracts. Plasmids containing cDNA inserts were excised from the recombinant lambda-Zap phage using Ex-assist helper phage and propogated in XLOLR cells.
