Organ: Root; Vector: pBK-CMV; Site_1: EcoRI; Site_2: XhoI; cDNA was synthesized from a pooled mRNA prep from elongating root hairs (30% w/w) and 2-3cm root tips (70% w/w). XhoI-oligo-dT linker-primer and EcoRI adaptors were used. cDNAs was cloned unidirectionally into lambda ZAP Express (Stratagene), amplified, and mass-excised into pBK-CMV vector plasmids.
