| Description |
Vector: pBluescript pSK; Site_1: EcoRI; Site_2: XhoI; Plants were grown in an aeroponic chamber for 14 days on nitrogen-rich medium followed by 3 days on N-free medium. RNA was extracted from root tips (1-3 cm). cDNA was prepared from polyA+ enriched RNA. The cDNA was directionally ligated into Uni-zapXR vector from Stratagene and packaged using Gigapack Gold packaging extracts. Plasmids containing cDNA inserts were mass-excised from phage stocks using ExAssit helper phage and propagated in SOLR cells. Clone ordering and sequencing was performed by the Centre National de Sequencage (Genoscope, Evry, France). |
