| Description |
Organ: stem; Vector: pSPORT 1; Site_1: SalI; Site_2: NotI; Plants were raised in growth chambers with a photoperiod of sixteen hours light and eight hours dark at 24oC and 50% humidity. At the start of the fourth-week following sowing, the outside of the stem above the second internode (counting up from the cotyledons) was scraped directly into a metal sieve over liquid N2. Total RNA was extracted using the Qiagen Maxi Total RNA kit, and polyA+ enriched RNA was obtained from this using Ambion Poly(A)Purist Mag. cDNA synthesis was primed using a NotI-oligodT primer, and using SalI adapters, double-stranded DNA was ligated into SalI NotI sites of pSPORT1. |
