| Description |
Vector: pDNR_lib (Clontech); Site_1: Sfi I A (GGCCATTACGGCC); Site_2: Sfi I B (GGCCGCCTCGGCC); Medicago truncatula A17 plants, were grown in pots containing 75% Metro Mix 350 soil and 25% sand. Glands were dislodged from tissues by vigorously vortexing tissues in an isotonic buffer. Glands were harvested by sieving out larger tissues and by differential centrifugation. RNA for library construction was isolated from pure gland preparations using the Qiagen RNA Easy kit. The Creator SMART cDNA Library Construction Kit from Clontech was used for library construction. |
