Description |
Vector: lambda-TriplEx2 (Clontech); Site_1: SfiIA; Site_2: SfiIB; The library HLUPJN1 was constructed from RNA isolated from lupulin glands from the variety 'Taurus'. Hop cones were collected near Mainburg, Germany in August 2002-2003. Lupulin glands were isolated by agitating dissected cones in liquid nitrogen followed by filtration to remove cone material. Total RNA was isolated using buffered phenol-chloroform-isoamyl alcohol followed by a clean-up with an RNeasy Plant Mini Kit (Qiagen); mRNA was purified using an Oligotex kit (Qiagen). The library was constructed using a SMART kit (Clontech). cDNAs were ligated into the phage vector lambda-TriplEx2 and packaged with MaxPlax Lambda packaging extract (Epicentre Technologies). For sequencing, cDNA inserts were PCR amplified from phage plaques using the vector primers 5'-CAAGCTCCGAGATCTGGACGAGC-3 and 5'-ATACGACTCACTATAGGGCGAATTGGCC-3'. |
