Vector: pYESTrp2; Site_1: EcoRI; Site_2: NotI; 3-4 weeks old seedlings were vacuum-infiltrated with a suspension of Pseudomonas syringae pv. tomato DC3000 hrcC minus strain (2x10E8cfu/ml) or left untreated. Leaf samples were collected 2h after treatment and mixed in a 20% untreated, 80% PAMP-elicited weight ratio before RNA extraction. cDNA first strand synthesis used an oligo dT (NotI) primer. cDNAs were directionally cloned into pYESTrp2 (Invitrogen). |
