RNA samples for cDNA library construction were prepared from six sources; roots from 3-month-old tomato (Solanum lycopersicum cv Micro-Tom) plants with flowers, roots from 3-month-old tomato plants without flowers, and Fusarium (F.oxyporum race2)-treated tomato roots (2-month-old) harvested 1 day and 1, 2, and 3weeks after infection. A full-length enriched cDNA library was constructed from a mixture of the RNAs by using a CAP-trapper method (Carninci and Hayashizaki, Methods Enzymol., 303: 19-44, 1999). |

Plasmids (pFLCIII, Carcini et al., Genomics 77: 79-90, 2001) prepared from randomly selected clones were used as templates for sequencing. 5'-end were sequenced using M13-21 primer and BigDye Terminator v3.1 Cycle Sequencing Kit (Applied Biosystems), and run on Applied Biosystems model 3730xl DNA Analyzer. |
