Vector: pDNR-LIB; Site_1: Sfi IA (GGCCATTACGGCC); Site_2: Sfi IB (GGCCGCCTCGGCC); Cannabis sativa plants were grown and processed in a DEA licensed and secured laboratory. Glands were removed from female inflorescences by vortexing, selected by sieving through a screen mesh, and collected by centrifugation. Total RNA was isolated from the glands using the Qiagen RNAeasy Plant Mini Kit (Qiagen Inc.-USA, Valencia, CA). The cDNA library from Cannabis trichomes was constructed using the Creator Smart cDNA library construction kit (Clontech, Palo Alto, CA, USA) following the manufacturer's protocol for total RNA. |
